Abstract: Celery yellow mosaic virus (CYMV), a member of potyvirus group, could be easily transmitted by mechanical inoculation, grafting and the aphid, Myzu.* Sulz . in a non circulative manner. It had a restricted host range , infecting only Umbelliferae and Chenopodiaceae plants.Symptoms commonly induced by CYMV in celery, coriander and parsley were chlorotic spots on the inoculated leaves and vein-clearing in young leaves. The infectivity of CYMV was lost: after 6 and 19 days in crude sap at room temperature (24-269C) and at 4-5’C, respectively; after diluting to 10““*, but not to 10“3; and after treating for 10 minutes at 50’C, but not at 45’C. Flexuous, elongated particles with a normal length of Ca. 760 nm were observed in leaf dips prepared from the CYMV-infected plants. Cytoplasmic lamellar inclusions and fibrous intranuclear inclusions were found in the CYMV-infected cells in ultra-thin sections. The intranuclear inclusions could be easily detected in epidermal strips with light microscope. This virus was successfully purified by clarification of the crude sap with 8% n-butanol, two cycles of differential centrifugation and one rate zonal density gradient centrifugation. An antiserum produced against the purified CYMV was specific and could be used to detect the virus in crude sap or purified preparation by double diffusion test in agar gel containg 0.5% of sodium dodecyl sulfate. The virus resembled western celery mosaic virus reported in North America in infectivity, transmission, morphology, physical properties in vitro , serological and cytopathological properties. However, some differences were found, mainly on the appearance of intranuclear inclusions and longevity in vitro. Under field conditions, the growth of 4 celery cultivars, mechanically inoculated with CYMV, was greatly affected in comparison to the non inoculated plants. Among these 4 cultivars, 'Florida 683 strain K’ and 'Tall Utah 52-70 strain H’ showed certain degree of tolerance. A survey made in the green belt of the Federal District showed that CYMV was commonly found in parsley and occasionally, in celery.
